Nadine wanted the new cells to light the plate the way the old cells had . A student was coming after lunch to borrow the promoter , and she had already told him it was ready : a piece of DNA he could take without a speech . She wanted that to be true before he walked through the door . The plate from the new host sat warm on the reader . Yesterday the wells had come up bright . These were dim . Same promoter , same reporter , a different cell , and a plasmid she had finished the night before , when the building was empty . She checked the blanks , then the labels , then the blanks again . Nothing had been swapped . The cells had grown . They were simply not reporting . She had been ready to blame a bad transformation , a dead batch , her own hands . That story was comfortable . A bad prep can be thrown out and started over . What she was looking at did not fit it . The part had worked in the first host . Around it , the sequence and the cell had changed , and the readout had changed with them . Marco stopped at the bench , saw her face , and did not ask if she was all right . He looked at the two printouts , the old plasmid and the new one , and tapped the short stretch where she had joined the promoter to the reporter . " You named the part , " he said . " The cell never did . " She waited for a fix . He took his coffee and left her with the plates . Nadine stayed with that longer than she liked . A promoter is where transcription starts . It is not a promise that transcription will give the same signal once the bases downstream are different , or once the host is . She had filed a measurement as if it belonged to the DNA alone . It had belonged to the DNA in that cell , in that join , on that day . She peeled the tape off the tube and wrote the host and the junction under the old name . When the student arrived , she did not hand him the tube . She showed him both plates and said he could use the promoter here , and that if he moved it he would have to measure it again . He nodded , a little disappointed , and the disappointment was the right size . She had wanted a part that traveled . What she had was a part that worked in one place , and she stopped calling those the same thing .
