En 1902 obtuvo una plaza de maestra en una escuela pública de Santander, ciudad en la cual trabajó brevemente. Después se trasladó a Bilbao, donde fue profesora en la academia anglo-francesa que había fundado su madre.
La Junta para Ampliación de Estudios, que buscaba modernizar la sociedad española mediante la educación, creó en 1915 el grupo femenino de la Residencia de Estudiantes. Ese grupo fue la Residencia de Señoritas, cuya directora fue María de Maeztu desde su apertura.
Maeztu estuvo al frente de la Residencia hasta 1936. El centro, que ofrecía alojamiento a estudiantes, también completaba su formación fuera de las aulas universitarias.
La Residencia tenía un laboratorio, en el cual las alumnas podían hacer prácticas, y una biblioteca, la cual servía de apoyo para sus estudios. También organizaba clases, conferencias, conciertos y lecturas de poesía.
El Instituto Internacional, el cual apoyó el proyecto desde Madrid, aportó edificios, profesorado y métodos educativos. Además, facilitó acuerdos con centros femeninos de Estados Unidos para ofrecer becas de intercambio, lo cual abrió nuevas oportunidades académicas.
Las residentes, quienes procedían de distintos lugares de España, compartieron aquel espacio con figuras como Victoria Kent, Matilde Huici, Delhy Tejero y Josefina Carabias. María Zambrano y Maruja Mallo formaron parte del profesorado.
Maeztu, cuyo trabajo también incluyó otras organizaciones, fundó el Lyceum Club Femenino. Este club, del cual fue la primera presidenta en 1926, nació en los salones de la Residencia de Señoritas.
Entre 1928 y 1930 formó parte de la Junta para Ampliación de Estudios, en la cual fue la única mujer antes de 1936. Allí participó como vocal.
En septiembre de 1936 renunció a la dirección de la Residencia, tras lo cual salió de España. Vivió en Argentina y murió en Mar del Plata en 1948.
